Shouldn't be a problem with a taxi - it's 2am when it is difficult when the world and his wife pile into the streets at club closing time. There is a taxi rank outside Victoria Station, where the MEN Arena is built over.
When the gig finishes, the buses and trams will still be running for a while longer, but as I say, tell me what area you are going to and I can answer more fully.

What's Manchester like on a Saturday night...I'm a southern softee who's got to walk from the Novotel on Dickinson Street to the MEN...I think it's about a mile. Am I likely to encounter any bother - if so where should I avoid?

Manchester is exactly the same as any other town or city on a Saturday night. Or any other night. One week you can be at a bus stop waiting and 4 dickheads will walk past and give you shit, the other week nothing will happen.
As long as you keep yourself to yourself and stick to the main roads you are fine.
If you want to make a scene and draw attention to yourself then you will get your just deserves.
Just keep an eye on what's going on around you and be aware of your surroundings and you will be OK.
